It won't absolutely prevent it, but it will make it far less likely. I use a kayak dry bag for the same reason.

Critters will still smell it. While the sealing will help, it won't keep mice away completely. Better to hang your food or sleep with it.

Hang it, never sleep with your food. Along a good portion of the AT you run the risk of bear encounters. Sleeping with your food doesn't protect it from a bear and may place yourself in danger. While bears along the AT are generally not a problem, all bets are off if food is involved.
