Your major road routes will lead you to Roan Mountain, Big Bald, Smokies, Hot Springs for the TN/NC sections. You can also head up to Virginia and do sections in the Mt Rogers area
If you head toward Boone using 421/321 & 19E, you can hit a number of trail heads Carvers Gap on Roan to Damascus, VA
If you go toward Asheville on I26, then you can either connect to Sams Gap & Erwin area.
Continue past Asheville on I40, you can go connect with the northern sections of the Smokies to Hot Springs. Then if you connect to US23 you can go into Franklin and the Nantalhalas.
If you want to do something easy, then I would go for Roan Mountain and north. If you want a good challenge then the Nantalhalas.
I-40W to 52N to 74 to I-77 to Burke's Garden in Va is probably the closest. Boone is two hours away and getting to Roan Mtn from Boone is a nightmare.

greensboro to the fox creek trailhead on the north side of the grayson highlands in approximately two hours.
I-40 west
421 north
I-77 north
highway 21 north near elkin (exits left off of I-77)
take highway 21 through sparta, nc
turn right on highway 93
at stop sign turn right to continue on 93 and cross into VA
at stop sign turn left onto highway 58
in volney continue straight onto highway 16
in troutdale turn left onto virginia 603.
fox creek trailhead is a few miles on the left
or, you could turn left to continue on 58 in volney instead of following 16 north to troutdale and grayson highlands state park will be a few miles down 58...
I believe the closest trail head would be at Atkins on I-81.
I-40 West to I-40 Business towards Winston Salem, just pass the Gboro airport
US 52 North (Mt Airy Exit)
I believe it is I-74 just South of Mt Airy that will run you out to I-77 North
I-77 to Wytheville where you will take I-81 South to Atkins
If you are lucky you can do it in under 2 hours.
Happy Hiking....
